Konstantin Konstantinov joined Ring as Chief Technology Officer in June 2023. Prior to joining Ring, Dr. Konstantinov served as the Executive Vice President of Manufacturing & Process Sciences and Chief Technology Officer at Codiak BioSciences. Prior to Codiak, he was the Vice President of Late-Stage Process Development responsible for bioprocess and technology development at Genzyme/Sanofi’s Boston Hub. Preceding Genzyme/Sanofi, Dr. Konstantinov held several positions of increasing responsibility at Bayer, concluding in the Head of Process Sciences. Over his extensive career, he has worked on the development and commercialization of various products and most recently pioneered the development of an end-to-end integrated continuous biomanufacturing platform and its application to traditional and new therapeutic modalities. He graduated with a bachelor’s in automation & control of continuous manufacturing and an MS in applied mathematics from Technical University of Sofia. Konstantin also received a PhD in biochemical engineering from Osaka University in Japan.

Dr. Konstantinov has published more than 60 peer reviewed papers and has over 15 patents and patent applications.

Keynote: Integration of Biology and Engineering to Enable Industrialization of Complex Biotherapeutics

This presentation addresses the challenges and opportunities associated with the development and scale-up of manufacturing processes for complex new biotherapeutic modalities. Beyond the recognized need for cutting-edge bioprocess engineering, we highlight the critical importance of industrialization-enabling biology, including manufacturable product structures, high potency, and optimized cell lines. Several examples of recent creative solutions will be presented. We will also review the latest bioprocessing methods and tools that, in synergy with the above mentioned biological advances, will deliver dramatically higher productivity, lower cost, and broader patient accessibility.

Dr. Zhenshu “Stan” Wang is a chemical engineer with a strong cross disciplinary foundation, earning triple majors in Chemical Engineering, Chemistry, and Mathematics from the University of Minnesota. He completed his MS and PhD at MIT under Prof. Yuriy Roman, with research centered on “Tuning geometric and electronic structure of noble metals using core–shell platforms to enhance catalytic performance.” Stan began his industry career at Eli Lilly in the Synthetic Molecule Design & Development organization as an Engineer Advisor, where he designed and optimized crystallization processes for both intermediates and the final API for Inluriyo (Imlunestrant). He subsequently joined Merck & Co. in Biologics Process Research & Development, working on downstream purification for monoclonal antibodies, fusion proteins, and antibody–drug conjugates (ADCs). At Merck, he employs process analytical technology (PAT) to improve process understanding and control, and he led an innovative collaboration with Levitronix^® to develop a single use TFF (tangential flow filtration) setup tailored for ADC processing. Across roles, Stan blends catalytic science, crystallization expertise, and biologics purification to deliver scalable, high quality manufacturing solutions.

Enabling a Single-Use TFF Platform for ADC Process Development

Antibody–drug conjugates (ADCs) demand downstream solutions that balance product integrity, operator safety, and process agility. We present a single‑use lab tangential flow filtration (TFF) platform purpose‑built for ADC process development, integrating Levitronix^® magnetically levitated pump technology and in‑line sensing to deliver gentle fluid handling, precise flow/pressure control, and rapid reconfiguration across different ADC programs. The platform employs disposable, closed flow paths to mitigate cross‑contamination risk and streamline setup/changeover, while minimizing holdup volume and exposure—critical for highly potent payloads.

Design considerations included shear reduction via centrifugal pumping and precise transmembrane pressure (TMP) control. Implementation allows for automatic UF and DF control with minimal human intervention. Operational metrics—setup time, leak points, and cleaning burden—also improved with standardized single‑use assemblies.

Case study spanning buffer exchange and concentration demonstrate reproducible performance from lab to pilot scales. Collectively, the Levitronix^®‑enabled single‑use TFF platform enhances robustness and safety while accelerating development timelines, offering a practical, cost saving solution for ADC purification that aligns with modern containment and tech‑transfer requirements. This work provides actionable design and control practices to facilitate broader adoption of single‑use TFF in ADC process development.

Johanna Wiesner works as a Principal Associate at Roche in the department of Gene Therapy Cell Culture Development in Penzberg, Germany. Forging the Link: A Strategy for Safe, Efficient and Robust Clarification at the Interface of USP and DSP in Viral Vector Production

As gene therapies rise in prominence, viral vectors have become preferred delivery vehicles for the gene of interest. Viral vectors can be used in gene therapies for the treatment of various diseases, such as ocular and neuromuscular disorders and diseases of the central nervous system.

The development of scalable manufacturing processes is crucial for delivering sufficient quantities of purified vectors for clinical and market supply. Usually, a substantial amount of development work is invested in upstream production and downstream purification, while the chain link connecting the two – the harvest – is often overlooked. This is evidenced by its limited representation in the available literature compared to the rest of the process. However, an optimized clarification is vital to maximize the viral vector yield of the upstream process, while simultaneously minimizing cell debris to enable efficient purification.

This presents a challenge, since the lysis required to release intracellular vectors results in a high burden of impurities. During early development at bench scale, centrifugation is commonly used to pellet cellular debris. However, this is difficult to scale up to a manufacturing scale, especially since single-use options are desirable for viral vector production as it reduces the risk of contamination of both product and personnel. Here, filtration emerges as a superior alternative that is available in a single-use format and easy to scale up. The developed clarification process showed that alluvial filtration is an efficient and robust method with >90% turbidity reduction and a viral vector recovery of >80%, which could successfully be scaled from bench scale to pilot and production scale. During the scale-up, compliance with strict German biosafety regulations (GenTSV) was paramount. The developed process ensures compliance through a fully closed, single-use, and automated system that prevents aerosol release, eliminates open handling steps and reduces risks of failure, while enabling continuous monitoring and documentation of vital process parameters.

Edward is the Sr. Technical Specialist in Cell Culture Pilot Plant at Roche/Genentech in South San Francisco, California. He has 22 years of experience focused on cell culture process development, technology evaluations, and the implementation of new technologies across the Roche/Genentech network, including high-turndown single-use bioreactors, a single-use harvest system/method, a single-use mixer, a cell retention device, single-use sensors, and a foam sensor. Edward holds multiple patents covering single-use bioreactors, methods of use for cell retention devices, and sensor technologies.

The Challenges to Scale Up Perfusion Process From Lab to Commercial Scale

As market demand for monoclonal antibody (mAb) biologics grows, optimizing and scaling bioprocessing techniques become crucial to boost productivity and fulfill patient needs. Process intensification, primarily through perfusion systems, provides an effective solution to the limitations of traditional fed-batch methods for low-productivity molecules. This method not only enhances the sustainability and efficiency of biomanufacturing but also proves to be cost-effective. The talk highlights collaboration with Levitronix to scale up the Levitronix single-use pump for perfusion processes from lab to commercial scale using the modeling tool. Whilestraightforward, it presents challenges due to the differing operating environments between lab and commercial scales.

Loïc Chappuis is a scientist at Merck Serono SA (an affiliate of Merck KGaA, Darmstadt, Germany) in the Process Technology Innovations group within the Global Drug Substances Development department. He has been working for 5 years on the development and optimization of the perfusion process platform. Loïc leads and supports different projects ranging from prototype testing to implementation at lab scale and tech transfer to pilot scale. Prior to this role Loïc contributed in the design and implementation of perfusion cell cultures at pilot and manufacturing scale for clinical material production. He holds a master’s degree in chemical engineering and biotechnology from EPFL.

Overcoming ATF Limitations in Large-Scale Perfusion: Assessment of HPTFF and rTFF Technologies

Long-term perfusion processes are increasingly used to intensify upstream operations, but robust implementation at large scale remains challenging. Alternating tangential flow filtration (ATF) has been the primary choice for cell retention due to its perceived ease of scalability. However, at manufacturing scale ATF can limit process performance due to filtration constraints, limited flexibility for further optimization, and high cost. In addition, our single-use upstream (USP) platform requires single-use cell retention devices that can be seamlessly connected to single-use bioreactors, while supporting harvest targets of up to 4000 L per day from a 2000 L perfusion bioreactor.

Our objective was to identify and characterize scalable, single-use–compatible, and cost-effective alternatives to ATF for cell retention in perfusion processes. We evaluated high-performance tangential flow filtration (HPTFF) and reverse tangential flow filtration (rTFF) across multiple scales. HPTFF was tested at 3.5–25 L working volume, while rTFF was assessed from 3.5 L up to 200 L. Performance was characterized in terms of cell retention efficiency, filtration behavior, ability to reach the required harvest rates, and impact on perfusion stability, product quantity, and product quality. In collaboration with Levitronix, we developed an rTFF cart at 200 L scale and operated it over several days to evaluate long-term robustness under intensified perfusion conditions.

Our results highlight the key opportunities and remaining challenges associated with HPTFF and rTFF as single-use–compatible alternatives to ATF.

Andrea is a Senior Scientist in the Bioprocess Technologies and Engineering Department at AstraZeneca. Prior to joining AstraZeneca, she received her Bachelor of Science in Chemical Engineering from Rensselaer Polytechnic Institute. In her time at AZ, she has focused on Tangential Flow Filtration (TFF) perfusion cell culture process development and evaluating novel technologies to improve filtration performance to enable a robust, scalable Integrated Dynamic Bioprocess (IDB).

Exploring the Impact of Reverse Tangential Flow Filtration (rTFF) Operation on Filter Performance and Fouling Mechanisms

With the growing demand for biologics, AstraZeneca has been investing in the development and scale-up of a dynamic perfusion cell culture process, utilizing a hollow fiber membrane for cell retention. A primary challenge with high cell density perfusion is membrane fouling, posing risks to process scalability and yield, due to increased filter transmembrane pressure, filter flux decay, and reduced product sieving. While Alternating Tangential Flow (ATF) has shown benefits in sustaining higher product sieving compared to Tangential Flow Filtration (TFF), ATF operation is challenged with high viscosity culture and lacks operational flexibility when scaling. This talk with detail studies performed in 3L perfusion bioreactors to assess Reversible Tangential Flow Filtration (rTFF), a novel cell retention operating mode, that leverages the benefit of alternating flow with improved process control and scalability. Various membrane types and rTFF alternating flow cycle times were assessed to understand the impact of operating mode (TFF vs. rTFF), cycle time, and membrane type on filter performance. Membrane staining methods were also employed to probe differences in fouling mechanisms between filter types and operating modes. The results highlight optimal rTFF operating parameters to improve filter performance, which can inform the design and scaling of filter technologies for perfusion cell culture.

His background includes extensive experience in containment strategies, process optimization, and the design of robust, scalable equipment solutions for high‑value manufacturing environments.

Paulo currently serves as Director of Process Equipment and Field Applications at ILC Dover. In this role, he supports Life Science companies in addressing complex challenges related to powder handling, containment, and in‑line mixing—helping manufacturers improve safety, efficiency, and overall process performance.

Enhancing Biopharmaceutical Production Through Advanced Mixing: Boosting Performance With EZJetFlo™ Technology

As the demand for monoclonal antibodies (mAbs) continues to grow across therapeutic, diagnostic, and industrial applications, biomanufacturers face mounting pressure to achieve higher productivity, increased process robustness, and improved scalability. Upstream operations—particularly media and buffer preparation, ingredient dissolution, and process homogeneity—play a critical role in ensuring consistent performance and maximizing overall yield. However, traditional mixing technologies often struggle to meet the evolving needs of intensified and flexible bioprocessing strategies while keeping operators and product safe. The EZJetFlo™ is an advanced in-line mixing system (based on the JetMixer) designed to overcome these upstream challenges by delivering rapid, energy‑efficient, and uniform mixing across vessels of varying scales. In her role, she leads the Viral Filtration operation and evaluates new technologies for implementation. She is also Digital Transformation lead for the Pilot Plant. Mackenzie holds a Bachelor of Science in Chemical Engineering from North Carolina State University.

Viral Filtration: Taking a Manual Process and Adding Control

Viral Filtration is a critical part of the viral clearance strategy in protein purification. This process, while simple, can require significant operator interaction to ensure critical operating conditions are maintained. At bench scale, pressure cans are typically used to drive liquid flow through a viral filter at a constant pressure. For larger scales, particularly in single use biomanufacturing facilities, where pumps are used to drive liquid flow, constant pressure can be difficult to maintain. When protein containing solution fills the filter, pressure increases rapidly if flow is not reduced. This process often requires the operator to manual change the flowrates rapidly to maintain operating pressure throughout the run. A single use Viral Filtration system from Levitronix^® was evaluated to introduce robust pressure control and process automation. Overall, the system simplifies and improves the operation, producing results that are consistent with benchtop development runs. This presentation will highlight the implementation of the Viral Filtration system at the pilot scale and show data comparing bench and pilot scale runs.

Julien Glibert is a P&AD Manager at Catalent Cell & Gene Therapy in Gosselies, Belgium, overseeing a team of scientists dedicated to advancing both upstream and downstream process development for pDNA manufacturing. His team works on integrated advanced technologies to improve throughput, product quality, and operational efficiency.

He received a master’s degree in Biochemistry and Molecular and Cell Biology in 2017 from the Université catholique de Louvain in Belgium. He subsequently worked at the university as a research associate focusing on inflammation before joining DelphiGenetics and later Catalent as a downstream scientist in process development for pDNA manufacturing.

In-Line Cell Lysis: A Scalable and Automated Approach to Bacterial Lysis

Efficient and robust plasmid DNA (pDNA) manufacturing is essential for cell and gene therapy applications. Lysis of E. coli cells stemming from the fermentation represents a critical step, aiming at recovering the plasmid material, and separating it from the rest of the cell components.

The In-Line Cell Lysis (ICL) system developed at Catalent Gosselies introduces a scalable, automated approach to bacterial lysis, leveraging advanced single-use assemblies and precise process control. By integrating Levitronix pumps, flowmeters, and mixers, the ICL system ensures optimal homogenization, accurate residence times, and reliable solution ratios, resulting in a streamlined workflow and consistent product quality.

Head Liposomal Formulation

Dr. Andreas Wagner is Head of Liposome Technology at Polymun Scientific GmbH. He has significant expertise formulation of liposomes and LNPs and development of the respective processes for their clinical use. He and the team at Polymun Scientific have significantly contributed to the 1^st successful mRNA vaccine Comirnaty by optimizing and up-scaling the LNP process as well as by supporting clinical and early market supply of the successful Covid-19 vaccine.

Dr Andreas Wagner studied Biotechnology at the University of Applied Life Sciences in Vienna, Austria and earned his Master and Ph.D. degrees in the field of biopharmaceutical technology. Dr Wagner is listed as inventor on multiple patents, like the liposome technology and some product patents of liposomal formulations. Furthermore, he has published several peer reviewed articles dealing with liposomes, the technology, products thereof and their application in preclinical and clinical studies.

LNP Process Development and Analytics for mRNA Vaccines and Targeted LNPs

Lipid nanoparticles (LNP) are the leading delivery systems for enabling the therapeutic potential of small interfering RNA (siRNA), mRNA for systemic applications or CRISPR. Lipid nanoparticles (LNPs), currently represent the most advanced platform for RNA delivery. The rapid development and approval of mRNA-based vaccines during the COVID-19 pandemic paved the way towards increased interest in mRNA LNP technologies. Besides IM administered vaccines, challenges like targeted delivery of LNPs to reach extrahepatic targets need to be solved. Efficient delivery of mRNA-lipid nanoparticles (LNPs) to specific cell types remains a major challenge for mRNA therapeutics. Conventional targeting approaches involve modifying the lipid composition or functionalizing the surface of LNPs, which complicates manufacturing.

Optimized LNP processes as well as a sound analytical method panel will be presented to show the feasibility of producing conventional as well as next generation LNPs, used in gene delivery or for extrahepatic therapeutic approaches.

Engineering Ice-Growth Geometry for Improved Cryopreservation of Protein and Cell Therapies

Decreasing temperature is generally expected to attenuate or arrest product degradation; however, freezing often introduces undesirable and unexpected quality issues. Freezing is inherently a process of microscale phase separation, in which solute concentrations increase dramatically as water crystallizes. This process is accompanied by pH shifts and elevated osmotic and hydrostatic pressures (reaching up to ~2 kbar), forcing concentrated phases and living cells through the porous ice network and exposing them to cold denaturation and significant mechanical stresses.

Here, we approach freeze–thaw engineering from the microscale to the bulk scale by computationally simulating local stresses experienced by proteins and cells using computational fluid dynamics. These simulations are used to design heat-transfer geometries that better control ice-growth behavior. Case studies demonstrate reduced protein aggregation and improved post-thaw functionality of stem cells, highlighting the importance of ice-growth geometry as a critical design parameter in cryopreservation systems.

Dr. Simone Blaesi is a highly experienced Validation Expert and Project Manager with over 12 years of expertise in the pharmaceutical industry, specializing in aseptic processing and bio-decontamination.

She currently serves as a Validation Expert at Roche within the MSAT Steriles department.In her current role, Simone leads the conceptualization and implementation of H2O2 bio-decontamination processes, ensuring global GMP and Annex 1 compliance. As a core member of global expert teams, she drives the harmonization of methodologies and has authored global strategy documents and SOPs. Her innovative work includes pioneering online process monitoring and successfully transitioning systems from formaldehyde to H2O2 technology.

Previously, Simone spent over six years at SKAN AG as a Project Manager Research and Process Engineer. There, she focused on wet-chemical H2O2 determination, method development, and providing expert consultancy for aseptic production.

Optimizing H2O2 Distribution in Material Transfer Chambers using Levitronix^® PURO™ Fan

In pharmaceutical manufacturing, ensuring the uniform distribution of vaporized hydrogen peroxide (VHP) during bio-decontamination is critical for maintaining sterile environments. Older designs of Material Transfer Chambers (MTCs) often face challenges with uneven gas distribution due to complex airflow patterns and varying load configurations. This study explores a technical solution to enhance bio-decontamination efficacy by integrating a Levitronix^® PURO™ fan—a high-purity, magnetically levitated fan system—directly into the chamber.
The evaluation focuses on comparing a standard MTC setup against a system modified with a PURO™ fan positioned at different locations.

To capture the operational extremes of the MTC, two distinct loads were analyzed.

The primary objective was to determine if the additional air circulation provided by the PURO™ fan could:
Homogenize the H2O2 concentration throughout the chamber volume.
Mitigate the impact of various load types on decontamination quality.
Enhance process reliability by reducing the “range” between maximum and minimum decontamination levels.

Testing involved a multi-variable approach, assessing different fan speeds, continuous versus pulsed operation modes, and varying decontamination durations. Results were monitored using both chemical indicators (CIs) to measure log-reduction ranges and biological indicators (BIs) to verify microbial inactivation. This presentation outlines the experimental setup and the methodology used to identify the optimal configuration for a more robust and load-independent bio-decontamination cycle.

Kamila is chemist and material scientist with PhD in physical chemistry from Jagiellonian University (Krakow, Poland). She has over 15 years of experience in realization and leading multidisciplinary R&D projects gained at Norwegian University of Science and Technology and SINTEF (Norway). Kamila is an author of over 50 scientific peer reviewed publications in the field of polymer chemistry, biophysics and material engineering.

She currently serves as R&D Project Manager at DEC Poland Tekpro where she leads innovative development projects, internally and externally financed, aiming at creating new products and improving existing technologies for powder handling and containment with a main focus on decontamination technologies.

Efficient Decontamination of Hard-to-Access Surfaces in Pharmaceutical Isolators Using Assisted Agent Distribution

Decontamination process of complex process equipment installed in pharmaceutical isolators can significantly benefit from forced mixing of a decontamination agent. Due to rigorous control of air-born particle concentration in clean environments such as pharmaceutical isolators, application of standard bearing-based fans to improve decontamination agent distribution is limited due to occurring abrasion and subsequent particle- contamination of the environment.

Here we present application of abrasion-free magnetic-levitation (MagLev) fan in decontamination of otherwise hardly accessible surfaces in a pharmaceutical isolator.

The model “hard-to-access surface” (HTAS) was set in the pharmaceutical isolator. The isolator was decontaminated using hydrogen peroxide in the form of an aerosol. Decontamination efficiency of the HTAS was tested using biological indicators with Geobacillus Stearothermophilus spores.

The case study presented here along with supporting Computational Fluid Dynamics modelling of air flow suggests that the decontamination efficiency of the HTAS is extremally dependent upon HTAS shape and the position of HTAS in respect to primary air stream induced by the fan as well as fan rotational speed and decontamination agent dose.

The results clearly suggest that appropriately designed assisted distribution system dedicated for pharmaceutical isolators ensures efficient decontamination of complex process equipment surfaces without the need of prolonging decontamination time.
